چکیده
Supplementary Information General Materials. The starting materials were all commercially available, and 1,4-bis(2hydroxyethoxy)-2-butyne (5b) and 2,4-hexadiyne-1,6-diol (8) were prepared according to the literature procedures. The cross-linking agents 3a, 3b, and 4b were reported previously, however, the physical and spectroscopic data of 4b were not described in the literature and were presented in this Supplementary Information.

Cross-link analysis of the C-telopeptide domain from type III collagen.
Several peptides were isolated from tryptic digests of insoluble calf aorta matrix by chromatography. Reductive pyridylethylation of a tryptic 15 kDa pool released fragments deriving from the C-terminus of type III collagen. A 50-residue peptide Tc(III) was shown by sequence analysis to be the C-terminal peptide from the alpha 1(III)-chain, containing a helical and non-helical region of equal s...
متن کاملAmino acid sequence of the triple-helical domain of human collagen type VI.
The complete amino acid sequence of the triple-helical domain of human collagen VI was deduced from sequences of appropriate cDNA clones and confirmed to about 50% by Edman degradation of tryptic peptides. This domain consists of three different peptide segments containing some 335-336 amino acid residues originating from central portions of the alpha 1 (VI), alpha 2(VI), and alpha 3(VI) chains...

In order to study the tendency of amino acid neighbors in helical structures, proteins with known structures were carefully analyzed. The studied helical positions: N , Ncap, N1, N2, N3, N4, M, C4, C3, C2, C1, Ccap, C and their doublet counterparts: N Ncap, NcapN1, N1N2, N2N3, N3N4, M1M2, M2M3, C4C3, C3C2, C2C1, C1Ccap, CcapC were carefully analyzed. The propensity for all amino acids i...
متن کاملAnalysis of secondary structure in proteins by chemical cross-linking coupled to MS.
Chemical cross-linking is an attractive technique for the study of the structure of protein complexes due to its low sample consumption and short analysis time. Furthermore, distance constraints obtained from the identification of cross-linked peptides by MS can be used to construct and validate protein models. If a sufficient number of distance constraints are obtained, then determining the se...
متن کاملInhibition of HIV type 1 infectivity by constrained alpha-helical peptides: implications for the viral fusion mechanism.
Linear peptides derived from the membrane proximal region of the gp41 ectodomain are effective inhibitors of HIV type 1 (HIV-1)-mediated fusion events. These inhibitory peptides lack structure in solution, rendering mechanistic interpretation of their activity difficult. Using structurally constrained analogs of these molecules, we demonstrate that the peptides inhibit infectivity by adopting a...
